Theobroma cacao L. es un cultivo de gran importancia por sus posibles aplicaciones en la farmacia y la agricultura. Contiene metabolitos que resultan de gran interés por sus funciones biológicas naturales en la respuesta de la planta a los diferentes tipos de estrés, tanto bióticos como abióticos. La presencia, tipo y concentración de los metabolitos (primarios y secundarios) pueden variar de acuerdo al genotipo analizado y se pueden acumular en estructuras asociadas a la arquitectura de la hoja como son los tricomas. El objetivo de este trabajo fue caracterizar 26 clones del banco de germoplasma de Padrón, INIA-Miranda, Estado Miranda, Venezuela y estudiar la relación existente entre los indicadores bioquímicos y el número de tricomas. Se determinó el contenido de compuestos fenólicos, proteínas solubles, prolina libre, carbohidratos totales, ligninas, el número de tricomas y la relación existente entre ellos. Los resultados mostraron que en el espacio bidimensional que explica el 46,68% de la variabilidad total, se definieron cinco grupos homogéneos: diez clones se asociaron con el máximo gradiente de prolina, carbohidratos totales y contenido de ligninas, nueve con alto contenido de proteínas y fenoles y siete con mayor número de tricomas en hojas. Se comprobó que el número de tricomas no se relacionó con la concentración de compuestos fenólicos. Los clones de cacao del tipo forastero mostraron mayor concentración de fenoles totales y carbohidratos, en tanto que los trinitarios presentaron mayor concentración de proteínas y prolina.


Theobroma cacao L. is a crop of major importance for their potential applications in pharmacy and agriculture. It contains metabolites that are of great interest for its natural biological functions in plant response to different types of stress, both biotic and abiotic. The presence, type and concentration of both primary and secondary metabolites may vary depending on the genotype analyzed and these compounds can accumulate in the architecture associated structures such as leaf trichomes. The aim of this study was to characterize 26 clones of the germplasm bank of Padrón, INIA-Miranda, Miranda State, Venezuela and study the relationship between biochemical indicators assessed and the number of trichomes. The content of phenolic compounds, soluble protein, free proline, total carbohydrates, lignins, the number of trichomes and the relationship between all traits was determined. The results showed that in the two dimensional space explained 46.68% of the variability, five homogeneous groups were defined: 10 clones were associated with the maximum gradient of proline, total carbohydrate and lignin content. Nine clones were associated with high content of phenols and a group of seven clones with the highest number of trichomes on leaves. It was found that the number of trichomes was not related to the concentration of phenolic compounds. The clones of Forastero cocoa type showed higher concentration of total phenols and carbohydrates, meanwhile the Trinitario type presented higher concentration of proteins and prolina.

Background: Cassava (Manihot esculenta Crantz) is a crop that is high in carbohydrates in the roots and in protein in the leaves, important for both human consumption and animal feed, and also has a significant industrial use for its starches. In this study we evaluated the genetic variability with molecular markers in different stages in micropropagated plants from somatic embryos of Venezuelan native clone 56. Results: Three markers were used: ISTR, AFLP and SSR, finding that ISTR showed the highest polymorphism among individuals tested. With AFLP a high similarity between the evaluated individuals was observed and with SSR total monomorphism was seen. Using cluster analysis it was found that individuals from an embryo labeled as fasciated at the beginning of the somatic embryogenesis process were grouped as independent of the other plants when analyzed at the acclimatization stage. The differences found with the different markers used are discussed. In field trials, micropropagated plants had a yield between 4 and 5 times the average yield of cassava in Venezuela. Conclusion: Despite variability in terms of DNA markers, somatic embryogenesis is suitable for mass propagation of highly performing cassava clones.

RESUMO

The systematic study of the genetic fingerprint (genomics) and the biochemistry (metabolites) that goes with a specific cellular process requires the characterization of all the small molecules that form the profile of metabolites and the associated genes. The metabolome represents the collection of all the metabolites during certain process in an organism. The transcriptome represents the gene expression profile, all the messengers RNA in a defined condition. Then to understand the whole process, the studies of metabolites must be accompanied with studies of the gene expression, hence the metabolome must be accompanied by the transcriptome, so we can identify genes and metabolites whose synthesis is induced by a specific process, an infection or stress. Studies of metabolomics generate an enormous amount of data, then they need mathematical and computational tools to establish the correlations between the biochemical and genetic data, and to build up networks that represent the complex metabolic interactions that occur in each case, using tools like Graph and Networks Theory to elucidate the emergent properties inherent to the complex interactions of the metabolic maps. This paper describes the major mathematical tools that can be used for these studies, with emphasis on a semi-qualitative proposal known as the kinetic structural model.

Amplified fragment-length polymorphism (AFLP) was used to evaluate the stability of DNA in regenerated plantlets of Coffea arabica obtained by direct (DSE) and indirect somatic embryogenesis (ISE). Cluster analysis using the unweighted pair-group method (UPGMA), showed no specific grouping pattern related to the type of embryogenesis. These results suggest that the somatic embryogenesis (SE) process has a mechanism for the selection of normal and competent cells. Bulked DNA from regenerated plants obtained by DSE and ISE, and from the mother plants, was used to characterize specific AFLP fragments associated with each SE process. Twenty-three primer combinations were tested. A total of 1446 bands were analyzed, with 11.4% being polymorphic and 84% being specific for regenerated plants. Furthermore, specific bands were detected for DSE, ISE, and the mother plants. These results indicate that the SE process induces rearrangements at the DNA level and demonstrates discrepancies between the mechanisms involved in each SE process. Coffea arabica breeding programs that involve DSE and ISE can use AFLP as an additional tool for assessing DNA stability.
